The Modern Family star posted a series of photos on Instagram. In the first photo, Ariel Winter stunned in the dark green blouse and black jeans. She continued to stun in the last two photos, which were shot from what looks like a hotel room. The actress encouraged her 4.6 million fans to look for the following items sitting behind her.

Cytosine Base Editor Enables Genome-Wide Genetic Function Screening | Genomeweb
NEW YORK – Using a cytosine base editor, researchers in China have developed a genome-wide gene function screening method, using CRISPR-based gene knockout that doesn't rely on double-stranded DNA breaks, or DSBs.
To solve this problem, they developed a DSB-independent, genome-wide CRISPR screening method, which they named iBARed cytosine base editing-mediated gene KO, or BARBEKO. The method uses cytosine base editors for genome-scale knockout screens by perturbing gene start codons or splice sites, or by introducing premature termination codons, and it's integrated with barcoded guide RNAs for improved screening quality and efficiency.
Using BARBEKO, the researchers constructed a cell library through lentiviral infection at a high multiplicity of infection, and achieved efficient and accurate screening results with a substantially reduced number of starting cells. Importantly, they found that in comparison with Cas9-mediated fitness screens, BARBEKO screens were not affected by DNA cleavage-induced cytotoxicity in HeLa-, K562-, or DSB-sensitive retinal pigmented epithelial 1 cells.
When they used BARBEKO in knockout experiments of gene function, the researchers found that the method yielded comprehensive lists of genes affecting cell proliferation, either positively or negatively. Importantly, they found that negative screening, which is usually more technically challenging, was much easier to accomplish with the new technique. In addition, gene-independent cytotoxicity triggered by Cas9-mediated cleavage often muddles the results of negative selection screens related to cell fitness, because the depletion level triggered by gene loss of function is generally modest — that was less of a problem with BARBEKO.
The researchers did note that newly optimized versions of cytosine base editors, or CBEs, were developed during the course of their research, with extended targeting scope via a flexible protospacer adjacent motif or an expanded activity window. This could be helpful to CBE-based library design with improved guide RNA quality and coverage, they added. About 1,700 genes are missing in the current version of the BARBEKO library because of the limited targeting scope of the CBE version they used. However, other CBE constructs with higher efficiency and fewer off-targeting in DNA and RNA could also be used.

Illinois Carjacker Returns Vehicle After Finding Toddler in Backseat
Even criminals can agree, parenting is way harder than prison. Which explains why a carjacker immediately returned to the scene of the crime after he discovered a toddler staring at him from the backseat.
It happened in Chicago’s West Side just around dinner time when a 26-year-old mother was forced from her SUV by an unknown assailant. The woman was out delivering food when her Nissan SUV was carjacked at a red light.
But the suspect didn’t even make it a block before turning around to return the three-year-old girl sitting in the back who demanded they sing ‘Baby Shark’ for the entirety of the joyride.
Carjackings have skyrocketed across the country since the start of the pandemic, with the number of incidents in Chicago doubling over the previous year. In 2020 alone America witnessed nearly 100,000 occurrences of shanghaiing a vehicle in motion.
It’s a warning to drivers everywhere to make sure your doors are locked at all times (and to slap a “baby on board” sticker to the back windshield to deter would-be highjackers).
While no suspects have been apprehended in this incident, the vehicle was later found abandoned, most likely because it smelled like crackers and apple sauce. With any luck, the person who committed this crime was scared straight after staring a toddler in the face and surviving. It’s a close call with parenting they’ll likely never forget.

Artist Ming Smith , in a recent interview with her gallerist Nicola Vassell, compared her photography technique to a shot in basketball: “The power of anticipation and the patience to wait for what’s coming. It’s like a basketball player hitting three-pointers: practice, repeat, practice, repeat. You get better, and still you’ll miss a few. In photography, you have to nail it the moment it’s in the lens. Take the shot when you see it.” In a project tied to the 25th season of the Women’s National Basketball Association, which launched last month at the Barclays Center in Brooklyn, Smith had the chance to shoot some of the players hitting those three-pointers.
Earlier this year, with Vassell’s help as creative director, Nike’s Jordan Brand brought on Smith to photograph women members of the Jordan Brand family who, together, represent the largest group of female athletes to be endorsed by Jordan. The photographs include portraits of Minnesota Lynx guard Crystal Dangerfield, New York Liberty guard Kia Nurse, Dallas Wings forward Satou Sabally, L.A. Sparks point guard Te’a Cooper, Dallas wings guard Chelsea Dungee, Los Angeles Sparks guard Arella Guirantes, Minnesota Lynx forward Aerial Powers, Las Vegas Aces forward Dearica Hamby, and Seattle Storm guard Jordin Canada.
Smith is known for the candid, impeccably composed photographs she has been making for over four decades, since she became the first and only female member of Kamoinge, a collective of Black photographers founded by Roy DeCarava. In her photographs for the Jordan shoot, the athletes are shown, both individually and as a group, standing in a field of tall grass, wearing black dresses or black pantsuits. In two of the photos, they flank six-time NBA champion and Charlotte Hornets owner Michael Jordan himself.
Of interacting with the athletes, Smith said she and the shoot’s stylist Carlos Nazario, global fashion director of i-D magazine, “were trying to capture their essence.”
She added, “Empowering images are not stereotypical. Our mothers didn’t have to show huge cleavage, but they were very female. They were women. We are women. So just their beauty is beautiful to me inside and out. It’s the physical and spiritual … the spirit.”
Earlier this spring, Vassell opened her new New York gallery with an exhibition of Smith’s photographs titled “Evidence.” A number of Smith’s images from the Jordan project will be on view at the gallery as the small pop-up exhibition from June 29 through July 2.
