Rapid and efficient COVID-19 testing has been a pressing concern since the onset of the pandemic. Various technologies have been employed in the detection of viral RNA or proteins, or the antibodies generated against them mucous or other easily collected bodily fluids. CRISPR-Cas technology has been similarly exploited to detect RNA extracted from nasopharyngeal swabs. The viral genes are first amplified by RT-LAMP before Cas12a binds with the complimentary ssRNA, leading to collateral cleavage of ssDNA fluorescent reporter molecules. This method takes only 40 minutes, compared with the several hours required by RT-qPCR, and necessitates less sophisticated equipment.
In a paper recently uploaded to the preprint server medRxiv * by Edward Málaga Trillo et al . (April 29 th , 2021), these methods are tested in combination with CRISPR-Cas detection in the field, demonstrating a sensitivity and specificity of 93.8% and 99%, respectively.
Viral genes N (934 bp) and E (540 bp), and the human gene POP7 (406 bp) were selected for RT-LAMP amplification, and the complimentary guide RNA-Cas12a enzyme to these genes were prepared. The reactions were allowed to run for ten minutes in the presence of 50 nM of the Cas12a-guide RNA complex.
Of the 276 samples, 28.3% were positive for the E gene while 29.3% were positive for the N gene, though two false positives and five false negatives were later detected. The limit of detection of the system was tested by intentional doping with synthetic RNA, finding that the RNA was detectable at only 5 viral copies per 10 µL of sample, 2 µL being the inactivation solution.
This study was performed in the context of widespread COVID-19 in Peru combined with a limited testing capacity and aims to provide a cost-effective method of clinical validation. The test methodology developed by the group termed rapid coronavirus sensitive monitoring from saliva (RCSMS), can easily be applied at both the primary care level and in diagnostic laboratories and could be an important tool in detecting SARS-CoV-2 in less developed countries in particular.

SARS-CoV-2 spike mutations during transmission among humans and to animals
In SARS-CoV, K479N and S487T were the mutations that enabled the receptor-binding domain (RBD) of the viral spike to bind to the human host receptor, angiotensin-converting enzyme 2 (ACE2).
On the other hand, they caused short though deadly outbreaks, in contrast to the SARS-CoV-2, which has already been circulating in humans for over a year. During this period, many mutations have emerged that appear to impact viral fitness, such as the D614G that rapidly became the dominant strain worldwide.
The UK variant of concern, B.1.1.1.7, has 17 new mutations, with eight being located in the spike. The South African (SA) variant B.1.351 also has many spike mutations. Similarly, many mutations have been reported during the human-animal transmission of the virus, during experimental passage and during human interactions with pet or farmed, or even zoo, animals.
The Q493 and N501 residues in SARS-CoV-2 correspond to the ACE2 contact residues K479N and S487T in SARS-CoV. The UK variant shows the mutation N501Y in the second of these, as well as delH69, delV70, delY145, A570D, P681H, T716I, S982A and D1118H.
The N501Y probably allows the spike to interact with the 41Y residue on the receptor via a pi-pi bond.
The SA variant, also called 501Y.V2, has the above mutation N501Y, in addition to D80A, D215G, K417N, E484K and A701V. Another variant has been identified in Africa that has the P681H mutation, like the UK variant. This is related to the presence of the furin cleavage site in the spike protein in SARS-CoV-2.
The N501Y mutation has shown positive selection in aged mice during successive viral passages, and is associated with improved replication. When passaged still further, two mutations, namely, Q493H and K417N, with higher pathogenicity, were selected. Interestingly, the Q493K is also associated with higher replication and pathogenesis in mice, while the second mutation, K471N, is present in the SA variant.
Yet another mouse variant that emerged after six passages has been found, which contains Q493K as well as a deletion between Q675 and N679.
The transmission to mink led to the emergence of the mutations Y453F, F486L and N501T, all in the RBD, the last of which probably improves RBD-ACE2 binding. It has been observed in the virus following its passage in ferrets, which, along with mink, have given rise to two mutations at the furin cleavage site.

"This Is Us" actor Justin Hartley and Moroccan actress Sofia Pernas are raising eyebrows after being spotted wearing what seemed like wedding bands during their latest outing in Los Angeles.
On Sunday, Page Six published exclusive snaps of Hartley, 44, and Pernas, 31, as they were relaxing on a Malibu beach. The two were all smiles as they sat on the sand and enjoyed some drinks together.
Hartley wore a dark blue V-neck shirt while Pernas rocked a stylish white top that showed off her shoulders and an ample amount of cleavage. Both sported ripped jeans and large sunglasses.
In one photo, the pair looked really happy as they were seen laughing together. In another snap, they had serious expressions on as Pernas planted a kiss on Hartley's left biceps.
Interestingly, Hartley and Pernas appeared to be wearing wedding bands on their left ring fingers in the photos. The actor had what seemed to be a red ring while the actress had a silver band.
Page Six reached out to the representatives for "The Young and The Restless" co-stars, but they did not comment on the issue.
If true, this would be Hartley's third marriage as he was previously married to Lindsay Korman from 2004 to 2012 and Chrishell Stause from 2017 to early 2021. On the other hand, this would be Pernas' first.
Hartley and Pernas first met on the set of "The Young and The Restless" when the latter joined the show in 2015. Stause also joined the show for a 10-episode arc after Pernas was done portraying her role in the series.
Hartley went on to marry Stause in 2017 while he and Pernas remained friends. In November 2019, Hartley shocked his fans when he filed for divorce after two years of marriage. He even indicated in his filing that their marriage ended on Nov. 22, 2019, as per Us Weekly .

Dr. Krumov: E. coli is not a secreting microorganism per se. For a protein to be secreted in E. coli , it is first secreted into the oxidizing periplasm where the necessary disulfide bridges are built. However, because the physical space of the periplasm is limiting, it restricts the overall titer of generated material.
Pichia can secrete soluble proteins into the culture supernatant. As a result, the produced protein is highly pure. No endotoxins are formed with Pichia, and there is no need for viral clearance!
