Planning Commission approves alternate mural for cannabis dispensary after previous
A Long Beach cannabis dispensary will have a less controversial mural painted on it after the city's Planning Commission approved an alternate design after residents of the Bluff Heights neighborhood objected to the "sexual" nature of an earlier design.
The original mural, approved by the city's Cultural Heritage Commission in February, showed a large female figure with a butterfly on her finger, but some residents were concerned with the amount of the female's cleavage shown in the rendering.
Since then, representatives from The Lift dispensary reached out to the community after objections to the original mural design were raised, and proposed an alternative design.

Michael Forrest, a representative from The Lift asked the Planning Commission to deny the original design in favor of the alternate, stating the shop wanted to "arrive at a design that better suits the neighborhood."
The new design approved by the Planning Commission Thursday is markedly different in both color and content. There is no woman on the design approved by the commission and the green-dominant design that was approved in February is now a mix of red, brown and green.
A separate purple-dominant mural that would have been installed on the west-facing wall of the dispensary was replaced with a plan to have much of that part of the structure painted a muted-purple color.
The couple who had appealed the decision to approve the original design in February did not attend the meeting.

As a consequence, even before the polls closed, Labour has been afire with hot takes about how it should respond: for if in 2020 Starmer appeared to take the party one step forward, in 2021 it has taken two steps back. Some have suggested a new leader is required or that Starmer must reinvent himself; Corbyn fans argue Labour needs to reiterate its support for the 2019 manifesto, while others have called for the former leader to be expelled. A number have said Starmer should forget about trying to unite young university graduates with older school leavers and just focus on the concerns of the former; then again, others say his emphasis should be the other way around. This confusing, contradictory set of responses is just more evidence of a divided party in existential crisis: one lacking the ability and confidence to see the wood for the trees.
If education has replaced class as the new divide, one factor remains constant with the old politics: the Conservatives usually know how to win elections while Labour struggles. Thursday was a confirmation of that. Labour has been appallingly bad at creating positive electoral coalitions that can deliver victory under first-past-the-post. That is why Labour's three periods of majority rule have all been preceded by the widespread sense that a prolonged period of Conservative rule had failed.
They each required some kind of crisis to undermine the legitimacy of the Conservatives as a governing party: 1945 came after Conservatives lost trust on domestic issues due to the war, 1964 by the Profumo scandal and 1997 by Black Wednesday. While many in the party lionise the genius of Clement Attlee, Harold Wilson and Tony Blair, they each needed the Conservatives to implode before they could enter No. 10.
He hoped he might not have to change much within the party to help it recover from the appalling 2019 defeat and those three election reverses which had preceded it: perhaps just not being Jeremy Corbyn would be enough? This is not to say Starmer did nothing. Making members face the implications of the Equality and Human Rights Commission report into antisemitism has not been easy; just saying Labour was patriotic created serious ructions; and accepting that Brexit had happened also caused the Labour leader internal big trouble. However, as last Thursday suggests, the pandemic has not highlighted – to many voters anyway – that Johnson's Conservative government has failed. It has not united the electorate behind Starmer's Labour: far from it.

Looking at the way in which various social groups have voted this time, we find that education, class and location were most likely to decide if the voter would choose the Mahajot or the NDA. This comes out from the Lokniti-CSDS post-poll survey.
On the basis of levels of education, there is a neat pattern: higher the access to education, greater the share of NDA votes in that group. In turn, the Mahajot could win a greater share of votes among those with no formal education and among those who had education only up to the primary level. In both these groups, the Mahajot outperformed the NDA. In terms of economic class too, a clear pattern emerges: The Mahajot being stronger among the poor, the NDA getting a major share of votes among middle and upper classes. However, among lower income groups, the share of both alliances was almost equal. The NDA has a huge lead (40 percentage points) over the Mahajot among urban voters and though the Mahajot polled well among rural voters, here too, the NDA still has not done badly either.
Lokniti-CSDS post-poll study shows a slight but significant advantage to the NDA among women compared to men. Age also shows an interesting pattern — the NDA seems less popular among the youngest voters — something we had noticed in 2016 also.

ZombiUnicorn calls out Facebook & Call of Duty over "sexism" from Ghost actor Jeff
Twitch streamer Natalie ‘ZombiUnicorn’ Casanova has called on Facebook Gaming and Call of Duty to take action over Jeff Leach, a partnered streamer on Facebook and voice actor of Ghost in the CoD series.
Jeff Leach was permanently banned from Twitch in 2017 after comments he made on a H1Z1 talk show about ZombiUnicorn, because Casanova had turned down an offer to be on the show when they discovered Leach was also a guest.
On May 7, another Facebook streamer, NitroLukeDX, posted a Twitter thread including clips of Leach talking about ZombiUnicorn, and other clips.

In contrast to crystallographic fragment screening experiments, compounds in repurposing libraries are chemically more complex (fig. S1A) ( 6 , 7 ). Thus, these compounds likely bind more specifically and with higher affinity ( 8 ). Because of the higher molecular weights, we performed cocrystallization experiments at a physiological pH of 7.5 instead of compound soaking into native crystals ( 9 ).
From the 5953 compounds in our screen, we obtained x-ray diffraction datasets for 2381 compounds, which we subjected to automated structure refinement followed by cluster analysis ( 10 ) and pan dataset density analysis (PanDDA) ( 11 ) (table S1). We observed additional electron density, indicating binding to M pro , for 43 compounds, which were classified as hits, representing 37 distinct compounds (tables S1, S2, and S3). From these, the binding mode could be unambiguously determined for 29 molecules ( Fig. 1A and table S4). The majority of hits were found in the active site of the enzyme. Of the 16 active site binders, six covalently bind as thioethers to Cys 145 , one compound binds covalently as a thiohemiacetal to Cys 145 , one is zinc-coordinated, and eight bind noncovalently. The remaining 13 compounds bind outside the active site at various locations ( Fig. 1A ).
Of the 43 hits from our x-ray screen, 37 compounds were available in quantities required for testing their antiviral activity against SARS-CoV-2 in cell assays (table S2). Nine compounds that reduced viral RNA (vRNA) replication by at least two orders of magnitude in Vero E6 cells (fig. S2) were further evaluated to determine the effective concentrations that reduced not only vRNA but also SARS-CoV-2 infectious particles by 50% (EC 50 ) ( Fig. 2 ). Additionally, AT7519 and ifenprodil, which showed slightly lower vRNA level reduction, were included because of their distinct binding sites outside of the active site. From these 11, seven compounds (AT7519, calpeptin, ifenprodil, MUT056399, pelitinib, tolperisone, and triglycidyl isocyanurate) exhibited a ≥100-fold reduction in infectious particles in combination with either a selectivity index [SI; calculated as the 50% cytotoxic concentration (CC 50 ) divided by the EC 50 ] of >5 or no cytotoxicity in the tested concentration range and are considered antivirally active (table S5).

Triglycidyl isocyanurate has antiviral activity (EC 50 = 30.02 μM, CC 50 > 100 μM) and adopts covalent and noncovalent binding modes to the active site. In both modes, the compound's central ring sits on top of the catalytic dyad (His 41 , Cys 145 ), and its three epoxypropyl substituents reach into subsites S1′, S1, and S2. The noncovalent binding mode is stabilized by hydrogen bonds to the main chain of Gly 143 and Gln 166 and to the side chain of His 163 . In the covalently bound form, one oxirane ring is opened by nucleophilic attack of Cys 145 , forming a thioether ( Fig. 3D ). Triglycidyl isocyanurate has been tested as an antitumor agent ( 16 ).
MUT056399 binds noncovalently to the active site (EC 50 = 38.24 μM, CC 50 > 100 μM). The diphenyl ether core of MUT056399 blocks access to the catalytic site, which consists of Cys 145 and His 41 . The terminal carboxamide group occupies pocket S1 and forms hydrogen bonds to the side chain of His 163 and the backbone of Phe 140 ( Fig. 3F ). The ethyl phenyl group of the molecule reaches deep into pocket S2, which is enlarged by a shift of the side chain of Met 49 out of the substrate binding pocket. MUT056399 was developed as an antibacterial agent against multidrug-resistant Staphylococcus aureus strains ( 21 ).
Quipazine maleate showed moderate antiviral activity (EC 50 = 31.64 μM, CC 50 > 100 μM). In the x-ray structure, only the maleate counterion is observed covalently bound as a thioether (supplementary text and fig. S3B). Maleate is observed in structures of six other compounds showing no antiviral activity. The observed antiviral activity is thus likely caused by an off-target effect of quipazine.
In general, the enzymatic activity of M pro relies on the architecture of the active site, which critically depends on the dimerization of the enzyme and the correct relative orientation of the subdomains. This could allow ligands that bind outside of the active site to affect activity. In fact, we identified two such allosteric binding sites of M pro .
Five compounds of our x-ray screen bind in a hydrophobic pocket in the C-terminal dimerization domain ( Fig. 4, A and B ), located close to the oxyanion hole in pocket S1 of the substrate binding site. One of these showed strong antiviral activity ( Fig. 2 ). Another compound binds between the catalytic and dimerization domains of M pro .
