The interaction between blood clotting and SARS-CoV-2 infection
Coagulopathy - a condition in which the blood's ability to clot is impaired, is recognized as a significant aspect of morbidity in COVID-19 patients. In a recent study, Prof Lewis Cantley and a team of researchers explored the role of coagulation factors in the SARS-CoV-2 infection. Their recent study is posted on the bioRxiv * server.
"Coagulation-induced cleavage enhances spike activation and increases viral entry into target cells."
They found that the proteases involved in clotting can also directly cleave the SARS-CoV-2 glycoprotein spike, enhancing the viral entry. They also found that protease inhibitors promiscuously inhibited the spike cleavage by both transmembrane serine proteases as well as coagulation factors. Spike cleavage by proteases is crucial to the viral entry; it facilitates membrane fusion and hijacks of the host cell by the viral genome.
Even as anti-SARS-CoV-2 vaccines are currently administered across the world, the continuing existence of a large reservoir of active cases over a long period will cause viral variants to emerge.
Because the coagulation cascade is orchestrated by a chain reaction of serine protease zymogens that are each activated by proteolytic processing, the researchers investigated if the proteases involved in the clotting also play a role in the SARS-CoV-2 spike cleavage.
They found that the coagulation factors, serine proteases factor Xa and thrombin, directly cleave SARS-CoV-2 spike, with more significant proteolytic activity against the SARS-CoV-2 peptide substrates than TMPRSS2. They also found that the S1/S2 boundary appears to be an even more optimal factor Xa substrate than peptide substrates derived from known physiological targets of factor Xa in coagulation.
They reported that the serine protease inhibitors suppress SARS-CoV-2 entry via inhibition of TMPRSS2. They observed that the TMPRSS2 is likely the primary cellular target of camostat and nafamostat.
Further, they found that the nafamostat is a versatile inhibitor of spike activation by various transmembrane serine proteases (TTSPs) and coagulation factors.
"We have mitigated the risk of artifact by using multiple orthogonal platforms, including VSV-based pseudovirus, HIV-based pseudovirus, and syncytia formation."

CVC Files Motion in Opposition to Broad Priority Motion | McDonnell Boehnen Hulbert & Berghoff
In its turn, Junior Party The University of California/Berkeley, the University of Vienna, and Emmanuelle Charpentier (collectively, "CVC") filed its motion in opposition to Senior Party The Broad Institute, Harvard University, and the Massachusetts Institute of Technology (collectively, "Broad") motion for priority in Interference No. 106,115. CVC's motion challenges Broad's priority claim and the bases Broad set forth therein, rebutting Broad's legal arguments and mentioning more than once that Jennifer Doudna and Emmanuelle Charpentier received the 2020 Nobel Prize in Chemistry (and by implication that the Broad inventors had not).
Marraffini recalls that CVC's disclosure of tracrRNA as a necessary component in the catalytic DNA cleavage complex "as well [as] the development of the [sgRNA]" were "the most important findings that the team made" because, before CVC's breakthrough, no one knew that tracrRNA was an active component in the complex. Before June 26, 2012, Zhang had no concept of a sgRNA CRISPR-Cas9 system of Count because he was unaware that tracrRNA was a necessary component of the catalytic CRISPR-Cas9 DNA-cleavage complex. . . . Prior to June 2012, the art taught that tracrRNA was merely an intermediate cofactor that RNase III and Cas9 needed for RNA processing to generate mature crRNA guides from long, unprocessed pre-crRNAs. Marraffini confirmed [this understanding in the art], testifying that tracrRNA was only "known . . . to be involved in the generation of guide RNAs." Marraffini also testified that, before June 26, 2012, he and Zhang only discussed "the importance of tracr for what we knew at the time was generation of RNA guides," and that "the system, as we knew it , required RNase III to generate guides." Q: So you . . . did you generally convey the view to Dr. Zhang that you thought the single-guide RNA would be an important tool for genome editing?
When asked if he communicated to Zhang that CVC's sgRNA CRISPR-Cas9 system would be an important tool for genome editing in eukaryotes specifically, Marraffini stated unequivocally, "yes":
Q : [W]hen you first conveyed to Dr. Zhang the single -- single-guide RNA system described in Jinek 2012, did you convey to him that you thought it would be an important tool for genome editing in eukaryotes specifically?
Based on this testimony, CVC argues that the evidence shows that Broad's inventors, and specifically Dr. Zhang, derived their claimed invention from the CVC inventors through Dr. Marraffini. The brief further characterizes (turnabout being fair play) their own litany of purported "failures" by the Zhang lab, particularly with regard to the "fallback position" set forth in Broad's priority brief, that work performed prior to CVC's March 2012 conception date should be considered by the Board in making its priority determination. This work, on alternative systems for gene editing ( e.g ., TALEN and ZFN), is not CRISPR, CVC argues, and without knowledge of CVC's sgRNA discovery the Broad inventors were pursuing (fruitlessly) more complicated versions of CRISPR that required, inter alia , RNAse III and unprocessed tracr and crRNA fragments. Indeed, CVC argues that until Dr. Marraffini's disclosure of its invention to Dr. Zhang there was no recognition by the Broad inventors of the role and importance of tracr RNA.
CVC sets out its own summary timeline of events by the parties, modifying the timeline presented in Broad's priority motion according to CVC's version of events as supported by the evidence set forth in this brief:
Turning to Broad's evidence of actual reduction to practice, CVC makes the argument that the evidence is uncorroborated, because three of the witnesses (Sanjana, Cong, and Cox) set forth in this table are named as inventors, citing Chen v. Bouchard , 347 F.3d 1299, 1309 (Fed. Cir. 2003):
The brief further alleges failure of proof, no contemporaneous inventor recognition, and failure of corroboration, for each of Broad's purported actual reductions to practice of eukaryotic CRISPR on July 20, July 28, July 31, and August, 2012, and further argues that because Broad did not establish the actual August date for this last instance of ARTP, the Board should consider it to be August 31, which is after CVC's August 9, 2012 asserted ARTP date.
An "UnLikely" Source of SARS-CoV-2 Remains a Possibility
Laboratory escape is described as an extremely unlikely source for SARS-CoV-2 in new WHO report, but questions posed to the researchers remain open.
Absent the finding of COVID-19 patient zero and identifying the initial SARS-CoV-2 vector, the recently reported World Health Organization (WHO) investigation into the possible origin in China could only narrow the possibilities within a range of likelihoods:
Questions about a possible laboratory origin and on how the WHO researchers inspected and assessed the laboratories were posed by several news outlets including France 24 , the DPA from Germany, and USA Today .To the first question at the briefing, which was on that topic, from a correspondent with the Telegraph in the UK,Peter Ben Embarek, PhD, a food scientist, program manager at the WHO, and co-lead of the investigation's international team, responded with both assurances and caveats.
"We've put it in our report. It has been assessed. And, of course, since this was not the key or main focus of the joint (with China) studies, it did not receive the same depth of attention and work as the other hypotheses," Embarek said.
"Not saying that it was impossible, but not the one we would start initially going deeper into and focusing our attention on," he elaborated."But this being said...if there is a need to further explore this and potentially other hypothesis, of course we will continue to look into this hypothesis."
In his closing remarks at the briefing, WHO Director-General, Tedros Adhanom Ghebreyesus, PhD, noted that while the team had visited several laboratories in Wuhan and had considered the possibility of a laboratory origination, they had encountered difficulties in accessing raw data in this and other areas of their investigation.
"I do not believe that this assessment was extensive enough," Ghebreyesus said."Although the team has concluded that a laboratory leak is the least likely hypothesis, this requires further investigation, potentially with additional missions involving specialist experts, which I am ready to deploy."
Researchers outside the WHO have presented evidence and rationale for ruling out the laboratory origin scenario, as well as in support of the possibility and the need for further investigation.
In a letter to the editor of Nature Medicine , Kristian Andersen, PhD, Department of Immunology and Microbiology, The Scripps Research Institute, La Jolla, California, and colleagues describe evaluating the SARS-CoV-2 genome and related viruses and finding no evidence of it having been bioengineered in a laboratory.

Of the 4960 nonduplicate studies identified, 4734 were excluded at title and abstract level. A total of 226 studies underwent full-text review. Of those, 61 were included in the systematic review, and 49 in the meta-analysis.
Quality of the studies was assessed using the Newcastle Ottawa Scale, designed for nonrandomized studies. Quality assessment also utilized the Outcome Reporting Bias in Trials II (ORBIT II) tool by applying it to the exposure groups in the observation studies in place of interventions from trials. The ORBIT II tool assesses the risk for outcome reporting bias, characterized as high/low/none for each outcome.
The Newcastle Ottawa Scale includes a total of nine potential points. Of the 61 studies included in the review, 87% (n=53) received seven or more points (high quality); the median score was eight (interquartile range, 7-9). Most of the studies (79%, n=48) included adjusted estimates. Risk for outcome reporting bias was identified as being generally low for the cardiovascular events (high risk in 6 studies), but high for the cardiovascular mortality outcome (high risk in 35 studies).
Limitations to the findings cited by the authors included incomplete reporting of outcomes and the possibility of outcome reporting bias, and poor precision in estimations of the risk for cardiovascular events for specific thresholds of both peptides.
"These limitations notwithstanding, our study provides new and potentially clinically useful information on the relationships of various thresholds of NT-proBNP and BNP levels with outcomes in patients with ESKD including increased risk for cardiovascular mortality, all-cause mortality, and cardiovascular events. Further work is warranted to identify how to incorporate the use of natriuretic peptides to stratify and reduce cardiovascular risk for patients with ESKD," the researchers said.
2021 Grammys looks: the best and worst - The Boar
T here is some commonality to us all: we love looking at the dazzling outfits of celebrities for red carpet events. Last month, we got to see them at the annual Grammy Award. As always, we got to judge whose outfits were breath-taking and which outfits should have stayed in the dressing room. Here are my takes on the 2021 Grammys looks; for better or for worse.
During the performance, she changed into a matching blazer and pinkish gogo boots. Finally, the singer graced us with her matching lingerie set for her last performance, staying faithful to her pink-sequin theme. All of her looks were signed Versace. According to me, the singer offered us the best styles and performances along with Megan Thee Stallion of the night.
Harry Styles is sliding into his role as a fashion icon. However, endorsing this type of role can have its ups and downs. This night was a mix between both. The red-carpet look, signed by Gucci, was an incredible source of debate on social media. His outfit was composed of brown trousers, a V-neck knit sweater, a plaid vest, a purple boa, and white boots. The plaid yellow vest was hugely commented on via Twitter, reminding everyone of the iconic outfit of Cher in Clueless.
While performing, he wore a full leather costume and a yellow boa. Some people think his outfits were a perfect representation of his styles of the moments (a reminder of his Vogue cover). Other thought that the boa, plaid and leather were a bit too much. Personally, I think it was pure genius and only Harry Styles could ‘rock’ those outfits.
From the moment I saw Bruno Mars during the Grammys I have been manifesting to go back in the 70s and have him as my prom date. This costume is not objectively fashionable but when it is Bruno Mars and Anderson Paak, we can make an exception. Signed Gucci for Paak and Ricky Regal for our favourite breakup song maker, those costumes were such opposites in colour as to perfectly match together. Black pants for each of them, sophisticated flower patterns on a coral shirt for Anderson Paak' and a black coat on flashy ruffles red shirt for Bruno Mars. During their performance, they each wore matching brunt orange retro suits to finalise their total night's look.
Not a lot to say here. Last year, Lizzo showed us incredible outfits and this year she was forgettable, which is rare of her. The two outfits were custom-made Balmain, two seaform dresses. The first was short and green and the second one, long and bubble-gum colour. I cannot stop comparing these looks to last year's and they are not on the same level. I hope next time Lizzo will be as incredible as she was in her previous outfits.
She started the night with an orange bold gown by Dolce & Gabbana, with a bow and diamond stance heels. The "savage" interpreter shocked us all during her first performance where she wore a white cabaret look with feathers. From this performance, she changed into a diamond bodysuit for our favourite badass song. Finally, Megan the Stallion really aimed for the best to make our jaw drop during the performance of our 2020 anthem, WAP, where she changed for a metallic bodysuit. Nothing to say except that those outfits can make up for the year.
I love her songs, but I wish I could style her. This Halloween style was in total discrepancy with everybody else's outfit. She wore a black dress with a skeleton embroidered on it. This outfit just did not reflect the styles we can see on her Instagram.
DaBaby started the night with a flashy pattern suit signed by Dolce & Gabbana on an orange turtleneck base. However, his outfit that clearly stood out, was his performance outfit: a white diamond encrusted suit by Chanel. The matching glove signed the entire look. It was an audacious choice of outfit which clearly paid off!
